Background:Premarin is a conjugated conjugated estrogen drug that is used to treat vaginal, vulvar, and anal itching caused by different human pathological processes. We sought to compare the clinical efficacy, cost effectiveness, and safety of Premarin for menopausal women with vulvar, vaginal, and anal itching. A multicenter, randomized, fixed-dose, two-period, open-label, prospective study of 12,882 patients was conducted in which 1,744 women were randomized to treatment or placebo. The primary outcome measure was the primary endpoint of oral Premarin (Premarin 0.4 mg) and the secondary outcome measure was clinical efficacy. There was a statistically significant reduction in oral Premarin compared with placebo in women who experienced clinical improvements (P <.05), and there was a statistically significant reduction in adverse events compared with those taking Premarin (P <.05). There was no clinically significant difference in adverse events. A significant reduction in oral Premarin was achieved with Premarin 0.4 mg compared with placebo with a difference in adverse events of 5.4 percent, and with Premarin 0.6 mg compared with placebo with a difference in adverse events of 3.2 percent, compared with a difference in adverse events of 5.3 percent, of which 2.8 percent were severe. No clinically significant difference in adverse events was observed between treatment and placebo groups, although adverse events were more common with Premarin than with placebo. The treatment group experienced clinical improvement, and there was a statistically significant reduction in clinical efficacy, and there was no significant difference in adverse events. In conclusion, Premarin is a highly effective treatment for vulvar, vaginal, and anal itching caused by different human pathological processes. There was a statistically significant reduction in oral Premarin compared with placebo in women who experienced clinical improvement, and there was a statistically significant reduction in adverse events compared with those taking Premarin.
